Abstract:

SUMMARY In this research, which was done for two years, we investigated various liquid and solid media without feeder ceells, for the ability to support growth of P. carinii. Data obtained from counting the organisms nuclei showed that, addition of Cycloheximide, penicillin, & streptomycin to neopeptoneN-Acetyle Glucoseamine (N.P.G. medium) can provides growth requirements of organism in its trophozoites form. We found that, preculture adjacence of P. carinii with group A streptococci or addition of some reducing substances, or vitamines to N.P.G. would increase the growth rate of organism. It was also shown that, addition of Neopeptone, N-Acetyl-Glucoseamine, and lowering its PH to 4-0, Rosewell Park Memorial Institute medium (R.P.M.I.), can also provides organisms growth. And also addition of agar & humans blood group Oto N.P.G. can enable P carinii to growth on the slant, and produce micro colonies.

Continuous axenic cultivation of Pneumocystis carinii.

Continuous axenic culture of Pneumocystis carinii has been achieved. A culture vessel is used that allows for frequent medium exchange without disturbance of organisms that grow attached to a collagen-coated porous membrane. The growth medium is based on Minimal Essential Medium with Earle's salt supplemented with S-adenosyl-L-methionine, putrescine, ferric pyrophosphate, N-acetyl glucosamine, ...

Activity of inosine analogs against Pneumocystis carinii in culture.

Three analogs of inosine, formycin B, allopurinol ribonucleoside, and 9-deazainosine, were tested for their ability to suppress proliferation of Pneumocystis carinii in culture with WI-38 cells. The organism was inhibited by 9-deazainosine at 10 micrograms/ml, and there was some inhibition at 1 microgram/ml. Formycin B was effective only at 40 micrograms/ml. Allopurinol ribonucleoside had littl...

Propagation of Pneumocystis carinii in Vero cell culture.

Pneumocystis carinii derived from infected murine lung was found to be capable of limited growth in Vero African green monkey kidney cell cultures. The observed increase in the number of cyst forms was influenced by the ratio of cysts to host cells in the inocula, duration of passage, and formulation of the cell culture media. Maximum growth was achieved by inoculating 1.3 X 10(5) cysts per 75-...

Pneumocystis carinii.

Although Giemsa staining has been routinely used for the detection of trophozoites and intracystic bodies in smears of bronchoalveolar lavage fluid (BAL) from patients with Pneumocystis cariniipneumonia, it does not normally stain the cyst wall. For detection ofthe cysts other stains such as toluidine Blue 'O' and methenamine silver must be used as well. Sulphation of smears before staining wit...

Biological profile and response to anti-pneumocystis agents of Pneumocystis carinii in cell culture.

Although the growth characteristics of Pneumocystis carinii have been described in several cell culture systems, the response of this organism to the drugs of choice, trimethoprim-sulfamethoxazole and pentamidine isethionate, have not been described in vitro. The effect of various concentrations of drugs against P. carinii on the growth of this potentially hazardous opportunistic organism as we...

Dictyostelium discoideum during Growth in Axenic Culture

1. The specific activities of fl-N-acetylglucosaminidase, acid phosphatase, ac-mannosidase, P-glucosidase, UDP-glucose pyrophosphorylase and alkaline phosphatase have been determined in myxamoebae of the cellular slime mould Dictyostelium discoideum Ax-2 grown on different media and in different phases of the growth cycle. 2. Variations in enzymic composition occur with changes in growth medium...
